What is the Germline 3 Gene Panel?

The Germline 3 Gene Panel evaluates BRCA1, BRCA2, and TP53—three critical genes linked to hereditary cancer syndromes. While BRCA1/BRCA2 mutations are associated with HBOC, TP53 mutations are linked to Li-Fraumeni syndrome, which increases the risk of multiple early-onset cancers, including sarcomas, breast cancer, brain tumors, and leukemia.

Indications for Testing

  • Patients with early-onset breast cancer or multiple primary cancers.
  • Individuals with a family history of BRCA1/BRCA2 or TP53-related cancers.
  • Those with a history of childhood sarcomas or brain tumors (suggestive of LFS).
  • To guide treatment decisions, including chemotherapy sensitivity (e.g., TP53 mutations may impact response) and  planning radiation therapy.

Inheritance Pattern

P/LP mutations in these genes follows autosomal dominant pattern of inheritance which lead to a significantly increased lifetime risk of cancer. First-degree relatives have a 50% chance of inheriting the mutation.
